I'm new to nVidia graphics drivers. What's the best method to install new drivers? Install right over the top of the old? Uninstall first? Use driver cleaner?

Pretty sure if you are using Windows 7 you are just as well off installing them over the top, because when you restart after uninstalling them Windows will install drivers from Windows Update automatically. Also, if you do not have any drivers installed in W7 the nvidia drivers will claim no hardware found.

I always uninstall, reboot, then install the new ones.

The problem is, sometimes Windows detects the card and loads it's own WHQL version, which requires another reboot because the Nvidia set up program detects another "install wizard" or something.
