This is my first card that i am attempting to overclock.
its an MSI 6600GT PCI express16

i go into MSI Clock and i have 3 settings

2D Clock set at 300MHz

3D Clock set at 500MHz

Memmory Clock set at 1000MHz

Which should i be ajusting and how much above Default

You don't need to touch your 2D (core) clock rate, only 3D. As one usually does when OC'ing, only go in small increments; i.e 5MHz or so at a time. Can't tell you what freq's you'll succesfully hit without any bugs though as I have no experience with a 6600 GT, maybe 580/1100 or higher, depending on voltages, cooling, BIOS version etc...

Don't forget to watch your temps too...

What jAkUp said, absolutely. Very important. Adding to that, pay close attention while running any benchies/stress tests (like DX9 RTHDRIBL) for any graphical glitches usually caused by an overheating GPU and/or VRAM, known as artifacts (loose black polys appearing randomly all over the screen is one common symptom of overheating).
